## Summary

AALL1521 is a single-arm phase 2 study run by Incyte with the Children's Oncology Group in children and young adults with newly diagnosed high-risk B-cell acute lymphoblastic leukaemia that is Philadelphia chromosome-like with a CRLF2 rearrangement or a JAK pathway mutation, or with other JAK-STAT activating lesions. Ruxolitinib was added to the AALL1131 high-risk chemotherapy backbone from consolidation onwards. Part 1 established the safe dose and schedule; Part 2 measures three-year event-free survival in each genetic cohort.

The study enrolled 171 patients and is recorded as completed with a primary completion date of March 2026. No primary results have been posted on the registry or published in a journal indexed by Europe PMC, so the corpus records what the trial tests and not a result.
