## TL;DR

The first trial to show that blocking a tumour's blood supply lengthens life. It added about five months and created a drug class.

## Summary

AVF2107g randomised previously untreated metastatic colorectal cancer to IFL chemotherapy with bevacizumab or placebo. Median overall survival was 20.3 against 15.6 months, hazard ratio for death 0.66. It was the first phase 3 trial of an anti-angiogenic antibody to show a survival benefit in any cancer and led to the FDA approval of bevacizumab in February 2004 and the EU marketing authorisation for Avastin on 12 January 2005. The chemotherapy backbone, bolus IFL, is obsolete and the size of the benefit shrank in later trials that used infusional regimens, which is why NICE TA1136 in February 2026 reached bevacizumab only as an option for first- and second-line use when targeted treatment or immunotherapy is unsuitable.
