## TL;DR

CheckMate 143 was the first large randomised test of an immune checkpoint drug in glioblastoma. When the tumour came back, nivolumab kept patients alive no longer than bevacizumab.

## Summary

CheckMate 143, trial NCT02017717 sponsored by BMS, randomised 369 patients with glioblastoma at first recurrence after radiotherapy and temozolomide, open-label, to nivolumab 3 mg/kg or bevacizumab 10 mg/kg every two weeks. Median overall survival, the primary endpoint, was 9.8 versus 10.0 months (HR 1.04, 95% CI 0.83-1.30), 12-month survival was 42% in both arms, and the objective response rate was higher with bevacizumab (23.1% versus 7.8%). Published by Reardon and colleagues in JAMA Oncology in 2020. It is one of three negative phase 3 trials of nivolumab in glioblastoma, with CheckMate 498 and CheckMate 548 in newly diagnosed disease.
