## Summary

CHRYSALIS is the phase 1 dose-escalation and expansion study of amivantamab, a bispecific antibody against EGFR and MET, in advanced non-small-cell lung cancer. Expansion cohorts tested the antibody alone in EGFR exon 20 insertion disease after platinum chemotherapy, and in combination with the third-generation EGFR inhibitor lazertinib or with chemotherapy in other EGFR-mutant settings.

The exon 20 insertion cohort, a group for which conventional EGFR inhibitors work poorly, showed a clinically meaningful response rate with durable responses, and amivantamab received accelerated approval in the United States in May 2021 for that indication. The combination cohorts with lazertinib provided the rationale for the phase 3 MARIPOSA and MARIPOSA-2 trials.
