## Summary

DNADalHR (NCT06970912) randomises 393 planned women with high-risk HR-positive, HER2-negative early breast cancer to four cycles of taxane-based chemotherapy or of dalpiciclib with an aromatase inhibitor before surgery; adjuvant treatment is then adjusted by ctDNA. The registry describes tumour-informed personalised ctDNA panels of 16 clonal variants from whole-exome sequencing. Primary endpoints are ctDNA clearance after neoadjuvant therapy and three-year event-free survival; primary completion is estimated for December 2029.
