## TL;DR

MAJIC-PV, a UK academic trial, found that ruxolitinib gave more complete responses than best available therapy and, for the first time, that a complete response went with fewer clots and progressions.

## Summary

MAJIC-PV randomised 180 patients with PV resistant to or intolerant of hydroxyurea to ruxolitinib or best available therapy. Complete response within a year, the primary endpoint, was reached by 43 percent on ruxolitinib versus 26 percent on best available therapy; complete responders had better event-free survival (thrombosis, haemorrhage, transformation or death), and the JAK2 V617F allele burden fell more on ruxolitinib (Harrison and colleagues, Journal of Clinical Oncology 2023). Registered as ISRCTN61925716.
