## TL;DR

The North American version of the same question, with the same answer: cetuximab after surgery adds toxicity and no benefit.

## Summary

N0147 randomised patients with resected stage III colon cancer to mFOLFOX6 with or without cetuximab. Three-year disease-free survival was 74.6 percent with chemotherapy alone against 71.5 percent with cetuximab added in KRAS wild-type disease (hazard ratio 1.21, 95% CI 0.98 to 1.49, p=0.08), and 67.1 against 65.0 percent in KRAS-mutant disease. Grade 3 or higher adverse events rose from 52.3 to 72.5 percent. No subgroup benefited. The trial also contributed the mismatch-repair and BRAF cohorts that later defined which stage III tumours are immunologically distinct, the biology that ATOMIC acted on thirteen years later.
