## TL;DR

The randomised trial behind CAPTEM: adding capecitabine to temozolomide lengthened the time before pancreatic neuroendocrine tumours grew.

## Summary

E2211, trial NCT01824875 run by the ECOG-ACRIN Cancer Research Group with the National Cancer Institute, randomised 144 patients with advanced pancreatic neuroendocrine tumours between April 2013 and March 2016 to temozolomide alone or capecitabine with temozolomide (CAPTEM). At the interim analysis in January 2018 the median progression-free survival was 14.4 months with temozolomide and 22.7 months with the combination (hazard ratio 0.58), which met the primary endpoint; at the final analysis in May 2021 overall survival did not differ significantly (53.8 vs 58.7 months, hazard ratio 0.82). MGMT deficiency was associated with response. ECOG-ACRIN ran the trial; the two medicines are generic.
