## Summary

OncoKB holds curated levels of evidence for the clinical actionability of somatic alterations, so that for a given mutation a reader can see whether an approved or investigational drug exists and how strong the evidence is. Memorial Sloan Kettering maintains it; it is free for academic use with a commercial licence, and was recognised by the FDA as a human variant database in 2021. Levels 1 to 4 grade therapeutic evidence and R1 and R2 mark resistance; an API is available and it is used by cBioPortal and many molecular tumour boards.
