## TL;DR

PLX-4545 is an experimental oral cereblon-based molecular glue degrader from Plexium, in phase 1 trials for solid tumours; its current status is not stated.

## Summary

PLX-4545 is an oral cereblon-based molecular glue degrader from Plexium. An oral molecular glue that degrades IKZF2 (Helios), a transcription factor that defines regulatory T cells; in healthy volunteers it shifted Tregs toward an effector-like phenotype. Preclinically combined with immune checkpoint therapy. Its furthest stage is phase 1, for solid tumours (preclinical, with immune checkpoint therapy). No clinical trial naming it is registered on ClinicalTrials.gov. First-in-human study in healthy volunteers reported at AACR 2025. PLX-4545 is not on Plexium's current pipeline page, which lists PLX-61639, PLX-66140 and an undisclosed degrader; no source found saying it was discontinued or partnered. It has not been shown to work in people.
