## Summary

PoMA sonodynamic therapy (PoMA) is a drug-device combination: oral 5-aminolevulinic acid activated by low-intensity defocused ultrasound (sonodynamic therapy) from Alpheus Medical. Oral 5-ALA is turned into protoporphyrin IX, which the company says builds up preferentially in malignant glioma cells; low-intensity ultrasound across the brain hemisphere then activates it to make reactive oxygen species that damage the tumour cells. A first-in-human study in recurrent high-grade glioma (NCT05362409, where the device is named CV01) is completed; a phase 2 with temozolomide in newly diagnosed glioblastoma (NCT07225621) and a phase 1 study NCT07862647 are recruiting.
