Treatment intensity was assigned by tumour biology (histology, MYCN, ploidy, 1p and 11q status) and response, with two to eight cycles of carboplatin, etoposide, cyclophosphamide and doxorubicin, surgery when resectable, isotretinoin for the highest-risk subset, and observation alone for some infants with stage 4S disease. The primary endpoint was three-year overall survival.\n\nThree-year event-free and overall survival were 83.2 and 94.9 percent, with overall survival of 100 percent for localised disease, despite reduced treatment for many subsets compared with earlier studies; the registry results give three-year overall survival of 99.4 percent in the lowest-intensity group and 88.4 percent in the highest. Infants with stage 4 disease and unfavourable biology did worse (three-year event-free survival 66.8 against 86.9 percent). The corpus's intermediate-risk neuroblastoma page cites ANBL0531 with A3961 for chemotherapy tailored by biology and response.","status":"positive","asOf":"2026-09-22","links":[{"label":"ClinicalTrials.gov NCT00499616","url":"https://clinicaltrials.gov/study/NCT00499616"}],"tags":["soc-trials"],"related":[],"cancers":["neuroblastoma-intermediate-risk","neuroblastoma","childhood-cancers"],"sections":[],"technologies":["cytotoxic-chemotherapy"],"targets":[],"drugs":["carboplatin","etoposide","cyclophosphamide","doxorubicin","topotecan","isotretinoin"],"companies":[],"institutions":["childrens-oncology-group"],"pathways":[],"terms":[],"trials":[],"people":[],"bottlenecks":[],"keyPapers":["paper-anbl0531-twist-jco-2019"],"journals":[],"dependsOn":[],"notes":[],"nct":"NCT00499616","phase":"3","setting":"Intermediate-risk neuroblastoma: response- and biology-based assignment to two, four or eight cycles of carboplatin, etoposide, cyclophosphamide and doxorubicin (with topotecan for poor responders) and surgery, with isotretinoin for the highest-risk subset and no chemotherapy for some stage 4S infants","sponsor":"Children's Oncology Group","result":"Three-year event-free survival 83.2 percent and overall survival 94.9 percent with response- and biology-based reduction of chemotherapy; overall survival 100 percent for localised disease.","yearReported":2019,"enrolled":464,"enrolledBasis":"registry","outcomes":[{"endpoint":"Overall survival at 3 years, all evaluable patients","primary":true,"unit":"%","arms":[{"name":"Response- and biology-based therapy","n":404,"value":94.9,"note":"95% CI 92.7 to 97.2"}],"source":"https://doi.org/10.1200/JCO.19.00919"},{"endpoint":"Event-free survival at 3 years, all evaluable patients","unit":"%","arms":[{"name":"Response- and biology-based therapy","n":404,"value":83.2,"note":"95% CI 79.4 to 87.0"}],"source":"https://doi.org/10.1200/JCO.19.00919"},{"endpoint":"Event-free survival at 3 years, infants with stage 4 disease by biology","unit":"%","arms":[{"name":"Favourable biology","n":61,"value":86.9},{"name":"One or more unfavourable biological features","n":47,"value":66.8}],"p":"0.02","source":"https://doi.org/10.1200/JCO.19.00919"},{"endpoint":"Overall survival at 3 years by treatment group (registry results)","primary":true,"unit":"%","arms":[{"name":"Group 2 (two cycles, surgery)","value":99.4},{"name":"Group 3 (four cycles, surgery)","value":93.5},{"name":"Group 4 (eight cycles, surgery, isotretinoin)","value":88.4}],"source":"https://clinicaltrials.gov/study/NCT00499616?tab=results"}]},"route":"/trials/anbl0531/","neighbours":{"cancer":[{"id":"childhood-cancers","kind":"cancer","name":"Childhood cancers (all types)","route":"/cancers/childhood-cancers/"},{"id":"neuroblastoma-intermediate-risk","kind":"cancer","name":"Intermediate-risk neuroblastoma","route":"/cancers/neuroblastoma-intermediate-risk/"},{"id":"neuroblastoma","kind":"cancer","name":"Neuroblastoma (paediatric)","route":"/cancers/neuroblastoma/"}],"technology":[{"id":"cytotoxic-chemotherapy","kind":"technology","name":"Cytotoxic chemotherapy","route":"/technologies/cytotoxic-chemotherapy/"}],"drug":[{"id":"carboplatin","kind":"drug","name":"Carboplatin","route":"/drugs/carboplatin/"},{"id":"cyclophosphamide","kind":"drug","name":"Cyclophosphamide","route":"/drugs/cyclophosphamide/"},{"id":"doxorubicin","kind":"drug","name":"Doxorubicin","route":"/drugs/doxorubicin/"},{"id":"etoposide","kind":"drug","name":"Etoposide","route":"/drugs/etoposide/"},{"id":"isotretinoin","kind":"drug","name":"Isotretinoin","route":"/drugs/isotretinoin/"},{"id":"topotecan","kind":"drug","name":"Topotecan","route":"/drugs/topotecan/"}],"institution":[{"id":"childrens-oncology-group","kind":"institution","name":"Children's Oncology Group (COG)","route":"/institutions/childrens-oncology-group/"}],"paper":[{"id":"paper-anbl0531-twist-jco-2019","kind":"paper","name":"ANBL0531: response- and biology-based therapy for intermediate-risk neuroblastoma","route":"/key-papers/paper-anbl0531-twist-jco-2019/"}]}}
