When the tumour came back, nivolumab kept patients alive no longer than bevacizumab.","summary":"CheckMate 143, trial NCT02017717 sponsored by BMS, randomised 369 patients with glioblastoma at first recurrence after radiotherapy and temozolomide, open-label, to nivolumab 3 mg/kg or bevacizumab 10 mg/kg every two weeks. Median overall survival, the primary endpoint, was 9.8 versus 10.0 months (HR 1.04, 95% CI 0.83-1.30), 12-month survival was 42% in both arms, and the objective response rate was higher with bevacizumab (23.1% versus 7.8%). Published by Reardon and colleagues in JAMA Oncology in 2020. It is one of three negative phase 3 trials of nivolumab in glioblastoma, with CheckMate 498 and CheckMate 548 in newly diagnosed disease.","status":"negative","asOf":"2026-09-17","links":[{"label":"ClinicalTrials.gov NCT02017717","url":"https://clinicaltrials.gov/study/NCT02017717"},{"label":"Reardon et al., Effect of nivolumab vs bevacizumab in patients with recurrent glioblastoma: the CheckMate 143 phase 3 randomized clinical trial (JAMA Oncology 2020)","url":"https://doi.org/10.1001/jamaoncol.2020.1024"}],"tags":[],"related":[],"cancers":["glioblastoma"],"sections":[],"technologies":["checkpoint-inhibitor"],"targets":["pd1"],"drugs":["nivolumab","bevacizumab-glioma"],"companies":[],"institutions":[],"pathways":[],"terms":["cold-vs-hot","blood-brain-barrier"],"trials":[],"people":[],"bottlenecks":[],"keyPapers":[],"journals":[],"dependsOn":[],"notes":[],"nct":"NCT02017717","phase":"3","setting":"Glioblastoma at first recurrence after radiotherapy and temozolomide: nivolumab vs bevacizumab","sponsor":"BMS","result":"OS 9.8 vs 10.0 months, HR 1.04 (95% CI 0.83-1.30); no benefit.","yearReported":2020,"enrolled":369,"outcomes":[{"endpoint":"Overall survival, first recurrence of glioblastoma","primary":true,"unit":"months","arms":[{"name":"Nivolumab","n":184,"value":9.8,"note":"Not significant"},{"name":"Bevacizumab","n":185,"value":10}],"hr":1.04,"ci":[0.83,1.3],"p":"0.76","source":"https://doi.org/10.1001/jamaoncol.2020.1024"}],"replication":"Consistent with CheckMate 498 and 548 in newly diagnosed disease: three negative randomised trials of PD-1 blockade in glioblastoma."},"route":"/trials/checkmate-143/","neighbours":{"cancer":[{"id":"glioblastoma","kind":"cancer","name":"Glioma & glioblastoma","route":"/cancers/glioblastoma/"}],"technology":[{"id":"checkpoint-inhibitor","kind":"technology","name":"Immune checkpoint inhibitors","route":"/technologies/checkpoint-inhibitor/"}],"target":[{"id":"pd1","kind":"target","name":"PD-1","route":"/targets/pd1/"}],"drug":[{"id":"bevacizumab-glioma","kind":"drug","name":"Bevacizumab (glioblastoma use)","route":"/drugs/bevacizumab-glioma/"},{"id":"nivolumab","kind":"drug","name":"Nivolumab","route":"/drugs/nivolumab/"}],"term":[{"id":"blood-brain-barrier","kind":"term","name":"Blood-brain barrier (BBB)","route":"/terms/blood-brain-barrier/"},{"id":"cold-vs-hot","kind":"term","name":"Hot vs cold tumours","route":"/terms/cold-vs-hot/"}],"idea":[{"id":"idea-neoadjuvant-io-glioblastoma","kind":"idea","name":"Neoadjuvant immunotherapy with surgical window for glioblastoma","route":"/ideas/idea-neoadjuvant-io-glioblastoma/"}]}}
