{"entity":{"id":"eclipse","kind":"trial","name":"ECLIPSE (GVAX pancreas and CRS-207)","aka":["ECLIPSE study","GVAX/CRS-207 phase 2b"],"tldr":"ECLIPSE was the randomised test of the Johns Hopkins pancreatic cancer vaccine strategy, a whole-cell GVAX vaccine followed by a Listeria vector carrying mesothelin; after promising early results it did not beat chemotherapy, with survival of 3.7 months against 4.6, and the programme ended.","summary":"ECLIPSE randomised 303 patients to cyclophosphamide with GVAX pancreas followed by CRS-207 (arm A), CRS-207 alone (arm B) or physician's choice single-agent chemotherapy (arm C). In the primary cohort of 213 patients who had failed two or more prior lines including gemcitabine, median overall survival was 3.7 months (95 percent confidence interval 2.9 to 5.3), 5.4 months (4.2 to 6.4) and 4.6 months (4.2 to 5.7) in arms A, B and C, with no difference between the vaccine combination and chemotherapy (hazard ratio 1.17, 0.84 to 1.64). Chills, pyrexia, fatigue and nausea were the commonest adverse events and there were no treatment-related deaths. The earlier phase 2 had reported 6.1 months with the combination against historical controls, a reminder that single-arm vaccine results in this disease have repeatedly failed to hold up when randomised, as ELI-002 7P did again in AMPLIFY-7P (2026); the personalised mRNA approach (autogene cevumeran) is now the vaccine strategy in randomised testing.","status":"negative","asOf":"2026-09-24","links":[{"label":"ClinicalTrials.gov NCT02004262","url":"https://clinicaltrials.gov/study/NCT02004262"},{"label":"Le et al., ECLIPSE (Clinical Cancer Research 2019)","url":"https://doi.org/10.1158/1078-0432.CCR-18-2992"}],"tags":[],"related":[],"cancers":["pancreatic","metastatic-pdac"],"sections":[],"technologies":["shared-antigen-vaccine"],"targets":["mesothelin"],"drugs":["cyclophosphamide"],"companies":[],"institutions":["johns-hopkins"],"pathways":[],"terms":["cold-vs-hot","pancreatic-failed-programmes"],"trials":["amplify-7p","autogene-cevumeran-phase-1"],"people":[],"bottlenecks":[],"keyPapers":[],"journals":[],"dependsOn":[],"notes":[],"nct":"NCT02004262","phase":"2","setting":"Previously treated metastatic pancreatic adenocarcinoma in the United States and Canada: cyclophosphamide with GVAX pancreas then CRS-207 (live attenuated Listeria expressing mesothelin), CRS-207 alone, or single-agent chemotherapy, randomised 1 to 1 to 1, with overall survival in patients after two or more prior lines as the primary endpoint","sponsor":"Aduro Biotech, with Johns Hopkins University","result":"Median overall survival 3.7 months with cyclophosphamide, GVAX pancreas and CRS-207 against 4.6 months with chemotherapy in the primary cohort (hazard ratio 1.17, 95 percent confidence interval 0.84 to 1.64); CRS-207 alone 5.4 months.","yearReported":2019,"enrolled":303,"enrolledBasis":"registry","outcomes":[{"endpoint":"Overall survival, primary cohort (2 or more prior lines)","primary":true,"unit":"months","arms":[{"name":"Cyclophosphamide + GVAX pancreas then CRS-207","value":3.7,"note":"95% CI 2.9 to 5.3"},{"name":"CRS-207 alone","value":5.4,"note":"95% CI 4.2 to 6.4"},{"name":"Single-agent chemotherapy","value":4.6,"note":"95% CI 4.2 to 5.7"}],"hr":1.17,"ci":[0.84,1.64],"source":"https://doi.org/10.1158/1078-0432.CCR-18-2992"}]},"route":"/trials/eclipse/","neighbours":{"cancer":[{"id":"metastatic-pdac","kind":"cancer","name":"Metastatic pancreatic ductal adenocarcinoma","route":"/cancers/metastatic-pdac/"},{"id":"pancreatic","kind":"cancer","name":"Pancreatic ductal adenocarcinoma","route":"/cancers/pancreatic/"}],"technology":[{"id":"shared-antigen-vaccine","kind":"technology","name":"Off-the-shelf cancer vaccines","route":"/technologies/shared-antigen-vaccine/"}],"target":[{"id":"mesothelin","kind":"target","name":"Mesothelin","route":"/targets/mesothelin/"}],"drug":[{"id":"cyclophosphamide","kind":"drug","name":"Cyclophosphamide","route":"/drugs/cyclophosphamide/"}],"institution":[{"id":"johns-hopkins","kind":"institution","name":"Johns Hopkins Hospital / Sidney Kimmel Comprehensive Cancer Center","route":"/institutions/johns-hopkins/"}],"term":[{"id":"cold-vs-hot","kind":"term","name":"Hot vs cold tumours","route":"/terms/cold-vs-hot/"},{"id":"pancreatic-failed-programmes","kind":"term","name":"Pancreatic cancer: the failed and stopped programmes and why","route":"/terms/pancreatic-failed-programmes/"}],"trial":[{"id":"amplify-7p","kind":"trial","name":"AMPLIFY-7P","route":"/trials/amplify-7p/"},{"id":"autogene-cevumeran-phase-1","kind":"trial","name":"Autogene cevumeran phase 1 in resected pancreatic cancer (Memorial Sloan Kettering)","route":"/trials/autogene-cevumeran-phase-1/"}]}}
