Patients were randomised to mogamulizumab, an afucosylated antibody against CCR4 that depletes malignant T cells, or to vorinostat, an approved histone deacetylase inhibitor; patients progressing on vorinostat could cross over.\n\nMogamulizumab significantly prolonged progression-free survival, the primary endpoint, with higher response rates, and the benefit was greatest in patients with blood involvement, the hallmark of Sezary syndrome. It was the first randomised trial in cutaneous T-cell lymphoma to compare two systemic drugs head to head and led to approval of mogamulizumab in the United States in August 2018.","status":"positive","asOf":"2026-09-17","links":[{"label":"ClinicalTrials.gov NCT01728805","url":"https://clinicaltrials.gov/study/NCT01728805"}],"tags":["subtype-trials"],"related":[],"cancers":["cutaneous-t-cell-lymphoma","peripheral-t-cell-lymphoma"],"sections":[],"technologies":[],"targets":[],"drugs":["mogamulizumab","vorinostat"],"companies":["kyowa-kirin"],"institutions":[],"pathways":[],"terms":[],"trials":[],"people":["youn-kim"],"bottlenecks":[],"keyPapers":["paper-mavoric-mogamulizumab-lancet-oncol-2018"],"journals":[],"dependsOn":[],"notes":[],"nct":"NCT01728805","phase":"3","setting":"Previously treated mycosis fungoides or Sezary syndrome: the anti-CCR4 antibody mogamulizumab against the oral HDAC inhibitor vorinostat, with crossover allowed","sponsor":"Kyowa Kirin","result":"Mogamulizumab improved progression-free survival over vorinostat in previously treated mycosis fungoides and Sezary syndrome; approved in August 2018.","yearReported":2018,"enrolled":372,"outcomes":[]},"route":"/trials/mavoric/","neighbours":{"cancer":[{"id":"cutaneous-t-cell-lymphoma","kind":"cancer","name":"Cutaneous T-cell lymphoma (mycosis fungoides and Sezary syndrome)","route":"/cancers/cutaneous-t-cell-lymphoma/"},{"id":"peripheral-t-cell-lymphoma","kind":"cancer","name":"Peripheral T-cell lymphomas (including cutaneous T-cell lymphoma)","route":"/cancers/peripheral-t-cell-lymphoma/"}],"drug":[{"id":"mogamulizumab","kind":"drug","name":"Mogamulizumab","route":"/drugs/mogamulizumab/"},{"id":"vorinostat","kind":"drug","name":"Vorinostat","route":"/drugs/vorinostat/"}],"company":[{"id":"kyowa-kirin","kind":"company","name":"Kyowa Kirin","route":"/companies/kyowa-kirin/"}],"person":[{"id":"youn-kim","kind":"person","name":"Youn H.
