The debt modern drugs owe that work is at the level of the pathway rather than the molecule, and the distinction is worth keeping: bortezomib and carfilzomib inhibit the proteasome, the protease at the end of the line, not the conjugation machinery the laureates characterised, and PROTACs and molecular glues were invented independently two decades later by exploiting the E1 to E3 cascade this work defined. The Nobel press release itself names no drug. Both laureates still publish from the Rappaport Faculty: Hershko from the Department of Biochemistry on how the anaphase-promoting complex is regulated, Ciechanover from the Rappaport-Technion Integrated Cancer Center on the ubiquitin-proteasome system in cancer and on degrader chemistry.
