He developed G47 delta, a triple-mutated third-generation oncolytic herpes simplex virus type 1.\n\nHis investigator-initiated single-arm phase 2 trial gave G47 delta repeatedly into the tumour in 19 adults with residual or recurrent supratentorial glioblastoma after radiotherapy and temozolomide. One-year survival after starting treatment was 84.2 per cent, the prespecified endpoint was met and the trial stopped early. Imaging showed a characteristic pattern of the lesion enlarging while contrast enhancement cleared after each dose, so conventional response criteria recorded almost every patient as stable disease, and biopsies showed rising CD4-positive and CD8-positive lymphocyte infiltration with persistently low regulatory T cells. The result led to the approval of G47 delta in Japan, the first oncolytic virus product approved there.","asOf":"2026-09-25","links":[{"label":"ORCID","url":"https://orcid.org/0000-0003-0523-8010"},{"label":"PubMed","url":"https://pubmed.ncbi.nlm.nih.gov/?term=Todo%20T%5BAuthor%5D%20oncolytic"},{"label":"Institute of Medical Science, The University of Tokyo","url":"https://www.ims.u-tokyo.ac.jp/imsut/en/"}],"tags":["oncolytic-virus","neuro-oncology","japan"],"related":[],"cancers":["glioblastoma"],"sections":[],"technologies":["oncolytic-virus"],"targets":[],"drugs":[],"companies":[],"institutions":[],"pathways":[],"terms":[],"trials":[],"people":[],"bottlenecks":[],"keyPapers":["paper-todo-g47delta-glioblastoma-natmed-2022"],"journals":[],"dependsOn":[],"notes":[],"role":"Professor, Division of Innovative Cancer Therapy, Advanced Clinical Research Center, and Department of Surgical Neuro-Oncology, Institute of Medical Science, The University of Tokyo","specialisms":["Oncolytic herpes simplex virus","Neuro-oncology","Glioblastoma","Investigator-initiated trials"],"profiles":[{"label":"ORCID","url":"https://orcid.org/0000-0003-0523-8010"},{"label":"PubMed","url":"https://pubmed.ncbi.nlm.nih.gov/?term=Todo%20T%5BAuthor%5D%20oncolytic"},{"label":"Institute of Medical Science, The University of Tokyo","url":"https://www.ims.u-tokyo.ac.jp/imsut/en/"}],"papers":[{"title":"Intratumoral oncolytic herpes virus G47 delta for residual or recurrent glioblastoma: a phase 2 trial","journal":"Nature Medicine","year":2022,"doi":"10.1038/s41591-022-01897-x"}],"orcid":"0000-0003-0523-8010"},"route":"/people/tomoki-todo/","neighbours":{"cancer":[{"id":"glioblastoma","kind":"cancer","name":"Glioma & glioblastoma","route":"/cancers/glioblastoma/"}],"technology":[{"id":"oncolytic-virus","kind":"technology","name":"Oncolytic viruses","route":"/technologies/oncolytic-virus/"}],"paper":[{"id":"paper-todo-g47delta-glioblastoma-natmed-2022","kind":"paper","name":"Todo 2022: the triple-mutated herpes virus that became Japan's first approved oncolytic virus","route":"/key-papers/paper-todo-g47delta-glioblastoma-natmed-2022/"}]}}
