ALX Oncology, based in South San Francisco, develops evorpacept (ALX148), an engineered SIRP-alpha fusion protein that blocks CD47 without the red-cell toxicity of earlier CD47 antibodies, so it can be combined with tumour-targeting antibodies to enhance macrophage attack. Its lead trial ASPEN-06 tests evorpacept with trastuzumab, ramucirumab and paclitaxel in previously treated HER2-positive gastric cancer, and the company is studying the drug in other antibody combinations.
