Eisbach Bio develops small-molecule cancer medicines. Its lead tablet, EIS-12656, blocks a DNA repair enzyme called ALC1 and is in an early trial in advanced solid tumours with defects in DNA repair.
Eisbach Bio GmbH is a privately held clinical-stage company in Martinsried, Planegg, near Munich. It uses an AI-guided platform called ALLOS to design small molecules that target the activated state of enzymes. EIS-12656 is an oral, brain-penetrant, allosteric inhibitor of the chromatin helicase ALC1 (CHD1L), studied in homologous recombination deficient tumours; the phase 1/2 MATCH trial (NCT06525298) at MD Anderson tests it alone and with olaparib or trastuzumab deruxtecan, the first patient was dosed in September 2024, and the trial is co-funded by the Cancer Focus Fund. Other programmes target CHD1 in PTEN-deficient tumours and WRN in MSI-high cancers. The company reported completing a phase 1 trial of the helicase inhibitor EIS-10700 in May 2024 and a CPRIT grant in November 2024. Its first registered cancer trial, NCT06525298, was posted on ClinicalTrials.gov in July 2024.
