Glycyx Therapeutics is testing whether axelopran, a pill that blocks opioid effects outside the brain, can help immunotherapy work in people who take opioids for cancer pain, without taking away their pain relief.
Glycyx Therapeutics develops axelopran, a peripherally acting mu-opioid receptor antagonist first developed for opioid-induced constipation, as an addition to PD-1 and PD-L1 checkpoint inhibitors in patients taking opioid analgesics. The company states axelopran has been given to 544 patients in earlier trials. Its first cancer trial, NCT07662720 (MORALE-HN01), a phase 1/2 study of axelopran with pembrolizumab in recurrent or metastatic head and neck squamous cell carcinoma, is registered with Glycyx as sponsor and NCI, Emory University and the University of Pittsburgh as collaborators. In fiscal year 2026 NCI awarded Glycyx MOR, Inc. USD 1,047,803 under SBIR award 5R44CA306565-02 for that trial.
