HotSpot Therapeutics targets natural regulatory hotspots, allosteric sites that control protein function, using its Smart Allostery platform. Its first clinical candidate HST-1011 is an oral allosteric inhibitor of the E3 ubiquitin ligase CBL-B, an intracellular checkpoint that limits T-cell and natural killer cell activation. HST-1011 is in a phase 1/2 trial alone and with cemiplimab in advanced solid tumours; the company also develops an IRF5 inhibitor for autoimmune disease.
