Oncopeptides is a Swedish company whose peptide-drug conjugate melflufen (Pepaxto) for relapsed multiple myeloma won accelerated US approval in 2021, was withdrawn there after a survival signal, and remains approved in Europe.
Overview
Oncopeptides, listed on Nasdaq Stockholm, developed melphalan flufenamide (melflufen, Pepaxto), a peptide-drug conjugate that concentrates an alkylating payload in cells rich in aminopeptidases such as myeloma cells. The FDA granted accelerated approval in February 2021 for heavily pretreated multiple myeloma, but the confirmatory OCEAN trial showed a possible overall survival detriment against pomalidomide and the company withdrew the US marketing authorisation; the European Commission approved Pepaxto in 2022 for a defined relapsed population and the company now markets it in Europe. It appears on OnCo as a lead sponsor of phase 3 cancer trials on ClinicalTrials.gov.
