Pasithea Therapeutics makes a tablet that blocks MEK, a switch in a growth signalling pathway. It is testing it in people with advanced cancers driven by that pathway and in adults with nerve tumours caused by neurofibromatosis type 1.
Pasithea Therapeutics Corp., of Miami Beach, Florida, was incorporated in Delaware on 12 May 2020 and listed on the Nasdaq Capital Market (KTTA) after its initial public offering in September 2021, according to its annual report on Form 10-K. It now focuses on RASopathies and MAPK pathway-driven tumours. Its lead candidate PAS-004 is an oral macrocyclic MEK1/2 inhibitor designed for a long half-life. A phase 1 trial (NCT06299839) tests PAS-004 in advanced solid tumours with RAS, NF1 or RAF mutations or after BRAF/MEK inhibitor failure, and a second phase 1 trial (NCT06961565) tests it in adults with NF1-associated plexiform neurofibromas. Its first registered cancer trial, NCT06299839, was posted on ClinicalTrials.gov in March 2024.
