Pathios Therapeutics makes PTT-4256, a tablet designed to stop the acidic surroundings of a tumour from switching off the immune cells inside it. It is in an early trial in people with advanced solid tumours.
Pathios Therapeutics Ltd is an Oxford, UK, drug discovery company established in 2017, according to the company. It targets GPR65, an acid-sensing G protein-coupled receptor on immune cells; the company's case is that the low pH of solid tumours signals through GPR65 to push T cells, NK cells, dendritic cells and especially macrophages towards an immunosuppressed, pro-tumour state. Its lead asset PTT-4256 is an orally bioavailable, selective GPR65 inhibitor. The company says it has raised a total of $58 million in venture funding, most recently a Series B to take PTT-4256 into the clinic. Its first registered cancer trial, NCT06634849 (RAISIC-1), a modular phase 1/2 dose-finding study of once-daily oral PTT-4256 in solid tumours at Australian sites, sponsored by its Australian subsidiary Pathios Therapeutics Pty Ltd, was posted on ClinicalTrials.gov in October 2024.
