Rhythm Pharmaceuticals develops treatments for rare diseases of the melanocortin-4 receptor pathway that controls hunger. Setmelanotide (Imcivree, RM-493) is approved in the United States and Europe for obesity due to POMC, PCSK1 or LEPR deficiency and for Bardet-Biedl syndrome. Its phase 3 TRANSCEND trial in acquired hypothalamic obesity, which follows surgery or radiotherapy for craniopharyngioma and other suprasellar tumours, met its primary endpoint in 2025 and supports a supplemental filing.
