SHY Therapeutics develops small-molecule medicines taken by mouth. Its lead, SHY-ONC6, blocks part of the proteasome, the cell's protein-recycling machine, and is in an early trial in advanced solid tumours.
SHY Therapeutics, LLC, of Valhalla, New York, with laboratory work on the New York Medical College campus, develops small molecules from a proprietary platform of pharmacophores that non-covalently modulate ATPases and GTPases. Its lead, SHY-ONC6, is described as an oral small-molecule inhibitor of proteasome ATPases; the company announced in August 2026 that the first patient had been dosed in its phase 1 trial, Luca-1 (NCT07705334), which gives SHY-ONC6 once daily in 21-day cycles to adults with advanced or metastatic solid tumours including triple-negative and HR-positive breast, colon and gastric cancers. Its pipeline also includes a multi-target Ras superfamily GTPase cancer programme in lead optimisation and non-oncology programmes in anti-infectives and neuromuscular disease. Its first registered cancer trial, NCT07705334, was posted on ClinicalTrials.gov in July 2026.
