TechnoGenesys makes (R)-9b, a tablet that blocks a protein called ACK1 which helps prostate cancer keep using male hormones after standard hormone drugs stop working. It is in a first trial in men with advanced prostate cancer.
TechnoGenesys, Inc., of St. Louis, Missouri, is commercialising (R)-9b, which it calls mahatinib, a small-molecule inhibitor of the androgen receptor-interacting tyrosine kinase ACK1 (TNK2). The company says ACK1 inhibition suppresses androgen receptor and AR-V7 expression, that (R)-9b resensitises enzalutamide- and abiraterone-resistant prostate cancer in preclinical work, and that it also promotes CD8 T-cell responses. The PHAROS phase 1 trial (NCT06705686, IND 167907) is the first clinical test of an ACK1 inhibitor, in metastatic castration-resistant prostate cancer. The company also names triple-negative breast and non-small cell lung cancer as later indications. Its first registered cancer trial, NCT06705686, was posted on ClinicalTrials.gov in November 2024.
