FGF1
FGF1 (Fibroblast growth factor 1) is a gene. The public catalogues list it as a drug target, and clinical evidence ties its variants to diagnosis, prognosis or drug response.
Overview
Plays an important role in the regulation of cell survival, cell division, angiogenesis, cell differentiation and cell migration. Functions as a potent mitogen in vitro. Acts as a ligand for FGFR1 and integrins.
Open Targets scores its association with cancer at 0.71 (direct and indirect evidence; datatypes literature 0.95, affected pathway 0.99, genetic association 0.04, clinical 0.65).

Biology
Plays an important role in the regulation of cell survival, cell division, angiogenesis, cell differentiation and cell migration. Functions as a potent mitogen in vitro. Acts as a ligand for FGFR1 and integrins. Binds to FGFR1 in the presence of heparin leading to FGFR1 dimerisation and activation via sequential autophosphorylation on tyrosine residues which act as docking sites for interacting proteins, leading to the activation of several signalling cascades. Binds to integrin ITGAV:ITGB3. Its binding to integrin, subsequent ternary complex formation with integrin and FGFR1, and the recruitment of PTPN11 to the complex are essential for FGF1 signalling. Location: Secreted; Cytoplasm; Cytoplasm, cell cortex; Cytoplasm, cytosol (UniProt). Locus 5q31.3 (HGNC).
