SHC1
SHC1 (SHC-transforming protein 1) is a gene. In the public catalogues the evidence so far is association rather than a proven role.
Overview
Signalling adapter that couples activated growth factor receptors to signalling pathways. Participates in a signalling cascade initiated by activated KIT and KITLG/SCF. Isoform p46Shc and isoform p52Shc, once phosphorylated, couple activated receptor tyrosine kinases to Ras via the recruitment of the GRB2/SOS complex and are implicated in the cytoplasmic propagation of mitogenic signals.
Open Targets scores its association with cancer at 0.66 (direct and indirect evidence; datatypes literature 0.97, affected pathway 0.84, animal model 0.49, genetic association 0.00).

No product in this corpus aims at SHC1 yet. Drugs bind the molecule precisely: to switch it off, flag the cell for the immune system, or deliver a payload.

Biology
Signalling adapter that couples activated growth factor receptors to signalling pathways. Participates in a signalling cascade initiated by activated KIT and KITLG/SCF. Isoform p46Shc and isoform p52Shc, once phosphorylated, couple activated receptor tyrosine kinases to Ras via the recruitment of the GRB2/SOS complex and are implicated in the cytoplasmic propagation of mitogenic signals. Isoform p46Shc and isoform p52Shc may thus function as initiators of the Ras signalling cascade in various non-neuronal systems. Isoform p66Shc does not mediate Ras activation, but is involved in signal transduction pathways that regulate the cellular response to oxidative stress and life span. Isoform p66Shc acts as a downstream target of the tumour suppressor p53 and is indispensable for the ability of stress-activated p53 to induce elevation of intracellular oxidants, cytochrome c release and apoptosis. Location: Cytoplasm; Cell junction, focal adhesion; Mitochondrion matrix; Mitochondrion (UniProt). Locus 1q21.3 (HGNC).
Notes
top- Written by scripts/fetch-cancer-genes.ts from CIViC, Open Targets, IntOGen, HGNC and UniProt; the function text is UniProt's, condensed and in UK spelling. Roles: none stated by the sources. Evidence tier "association-only" is the strongest of those signals.
- Prevalence not recorded: none of the sources gives a positivity rate.
