DESTINY-Gastric02 repeated the trastuzumab deruxtecan study in patients from Europe and the United States, after the first trial had been run only in Japan and Korea, and confirmed that the drug produces meaningful responses in HER2-positive stomach cancer after trastuzumab.
Overview
DESTINY-Gastric01 enrolled only East Asian patients, where stomach cancer is treated differently and diagnosed earlier. DESTINY-Gastric02 was a single-arm phase 2 study in the United States and Europe of trastuzumab deruxtecan in 79 patients with HER2-positive gastric or gastro-oesophageal junction cancer that had progressed on a first-line trastuzumab regimen, with the HER2 status re-confirmed on a fresh biopsy. The primary endpoint was confirmed objective response by independent review.
The study confirmed clinically meaningful and durable responses in a Western population, with a safety profile consistent with the earlier trial, supporting the drug's use outside East Asia. The subtype page notes that the response rate was lower than in the Asian trial.
