One-carbon Therapeutics develops medicines that exploit cancer cells' dependence on a metabolic pathway used to build DNA. Its lead medicine, TH9619, is in an early trial in advanced solid tumours.
One-carbon Therapeutics AB is headquartered in Solna, Sweden, and was founded as a spin-out from Karolinska Institutet; the company names Professor Thomas Helleday as its founder. TH9619 is a first-in-class small-molecule dual inhibitor of MTHFD1 and MTHFD2, enzymes of one-carbon metabolism, with MTHFD2 described by the company as highly overexpressed and cancer-specific. The first-in-human phase 1/2 ODIN trial (NCT07151040) in advanced solid tumours, including colorectal, head and neck, lung, gastric and gastro-oesophageal junction cancers, dosed its first subject in November 2025. In December 2025 the company closed an oversubscribed private placement of 153 million Swedish kronor (13.9 million euros), and in March 2026 it announced a collaboration with Tempus on TH9619. Its first registered cancer trial, NCT07151040, was posted on ClinicalTrials.gov in September 2025.
