Xilio Therapeutics
Xilio designs immune-stimulating drugs, such as interleukin-2 and checkpoint antibodies, with a molecular mask that is removed only inside tumours, so they act where the cancer is and spare the rest of the body. Its newest drug combines a PD-1 antibody with masked IL-2 and has just been cleared to start trials.
Overview
Xilio Therapeutics develops tumour-selective, masked immuno-oncology therapies. Clinical assets include vilastobart (masked anti-CTLA-4) and efarindodekin alfa (masked IL-12). In August 2026 the FDA cleared the IND for XTX501, a bispecific PD-1 / masked IL-2 designed to stimulate PD-1-positive antigen-experienced T cells while avoiding IL-2 receptor-mediated clearance and peripheral toxicity; a phase 1/2 trial will begin in metastatic non-small cell lung cancer and selected advanced solid tumours. IND-enabling work is under way on multi-specific masked T-cell engagers targeting CLDN18.2 and PSMA plus STEAP1, with INDs expected in the second half of 2027. Xilio is based in Waltham, Massachusetts, trades on Nasdaq as XLO and reported cash runway into the first quarter of 2028.
