CTO1681 is a tablet from CytoAgents meant to prevent and treat cytokine release syndrome, an immune over-reaction that can follow CAR-T cell therapy for blood cancers.
CTO1681 is CytoAgents' oral anti-inflammatory compound. ClinicalTrials.gov lists NCT05905328, a phase 1/2 study sponsored by CytoAgents of CTO1681 for the prevention and treatment of cytokine release syndrome in patients receiving CAR T-cell therapy, as recruiting. A 2026 National Cancer Institute SBIR award to CytoAgents extends the work to people with multiple myeloma receiving CAR T-cell therapy. It is a supportive medicine: it is not meant to treat the cancer itself.
Described by CytoAgents as a steroid-sparing inhibitor of prostaglandin and prostacyclin-mediated inflammation, given by mouth to damp the cytokine surge of cytokine release syndrome.
