MAGNOLIA: zanubrutinib in relapsed or refractory marginal zone lymphoma
The BTK inhibitor zanubrutinib produced responses in about two thirds of patients with relapsed marginal zone lymphoma across all subtypes with few of the cardiac side effects seen with ibrutinib, leading to its approval.
Overview
Phase 2 study of 68 patients with relapsed or refractory marginal zone lymphoma after at least one anti-CD20-based regimen treated with zanubrutinib 160 mg twice daily.
Objective response by independent review was 68 percent with complete response in 26 percent, similar across extranodal, nodal and splenic subtypes, with 15-month progression-free survival of 83 percent and low rates of atrial fibrillation or major bleeding.
