AUGMENT
AUGMENT showed that pairing rituximab with the immune-modulating tablet lenalidomide kept relapsed follicular lymphoma under control for much longer than rituximab alone, giving patients a chemotherapy-free option.
Overview
AUGMENT randomised 358 patients with relapsed or refractory follicular lymphoma (grades 1 to 3a) or marginal zone lymphoma who were not rituximab-refractory to rituximab plus lenalidomide for 12 cycles or rituximab plus placebo. The primary endpoint was progression-free survival by independent review.
The lenalidomide combination roughly tripled median progression-free survival compared with rituximab alone and improved response rates, at the cost of more neutropenia and skin reactions. It was approved in the United States in May 2019 for previously treated follicular and marginal zone lymphoma and became the reference chemotherapy-free regimen against which bispecific antibodies are now being tested.
