EGR2
EGR2 (E3 SUMO-protein ligase EGR2) is a gene that drives cell growth when it is altered. The public catalogues list it as an oncogene driver, and it is called a cancer driver by mutation analysis of patient cohorts. Tied to Chronic lymphocytic leukaemia.
Overview
Sequence-specific DNA-binding transcription factor. Plays a role in hindbrain segmentation by regulating the expression of a subset of homeobox containing genes and in Schwann cell myelination by regulating the expression of genes involved in the formation and maintenance of myelin. Binds to two EGR2-consensus sites EGR2A (5'-CTGTAGGAG-3') and EGR2B (5'-ATGTAGGTG-3') in the HOXB3 enhancer and promotes HOXB3 transcriptional activation.
IntOGen calls it a driver in 1 cohort (1 activating, 0 loss-of-function), covering Chronic Lymphocytic Leukaemia/Small Lymphocytic Lymphoma.

Sequence-specific DNA-binding transcription factor.

No product in this corpus aims at EGR2 yet. Drugs fit a pocket that exists only in one shape of the mutant protein and hold it there, off.

Biology
Sequence-specific DNA-binding transcription factor. Plays a role in hindbrain segmentation by regulating the expression of a subset of homeobox containing genes and in Schwann cell myelination by regulating the expression of genes involved in the formation and maintenance of myelin. Binds to two EGR2-consensus sites EGR2A (5'-CTGTAGGAG-3') and EGR2B (5'-ATGTAGGTG-3') in the HOXB3 enhancer and promotes HOXB3 transcriptional activation. Binds to specific DNA sites located in the promoter region of HOXA4, HOXB2 and ERBB2. Regulates hindbrain segmentation by controlling the expression of Hox genes, such as HOXA4, HOXB3 and HOXB2, and thereby specifying odd and even rhombomeres. Promotes the expression of HOXB3 in the rhombomere r5 in the hindbrain. Location: Nucleus (UniProt). Locus 10q21.3 (HGNC).

Notes
top- Written by scripts/fetch-cancer-genes.ts from CIViC, Open Targets, IntOGen, HGNC and UniProt; the function text is UniProt's, condensed and in UK spelling. Roles: IntOGen calls it an activating (Act) driver in 1 cohort. Evidence tier "cohort-driver" is the strongest of those signals.
- Prevalence not recorded: none of the sources gives a positivity rate.
