CATNON (EORTC 26053-22054)
CATNON showed that a year of temozolomide after radiotherapy lengthens survival in anaplastic gliomas that lack the 1p/19q co-deletion, and that the benefit is concentrated in IDH-mutant tumours.
Overview
CATNON randomised 751 adults with newly diagnosed 1p/19q non-co-deleted anaplastic glioma in a two-by-two design to radiotherapy alone, with concurrent temozolomide, with 12 cycles of adjuvant temozolomide, or both. Adjuvant temozolomide improved overall survival (hazard ratio 0.65 in the interim analysis; five-year survival 55.9 vs 44.1 percent), while concurrent temozolomide did not in the tumours as a whole and helped only IDH-mutant tumours (van den Bent and colleagues, Lancet 2017 and Lancet Oncology 2021).
