PHOENIX, a UK trial from the Institute of Cancer Research, gives women whose triple-negative breast cancer has not responded fully to pre-operative chemotherapy a short course of a DNA-repair drug (olaparib or ceralasertib) or immunotherapy (durvalumab) before surgery, and measures what it does to the tumour, to find which patients could benefit from these drugs after surgery.
PHOENIX (NCT03740893) is a non-randomised, biomarker-driven phase 2 window-of-opportunity and adjuvant study for 119 patients with neoadjuvant-treatment-resistant residual triple-negative breast cancer, run by the Institute of Cancer Research with AstraZeneca. Cohorts receive olaparib, the ATR inhibitor ceralasertib (AZD6738) or durvalumab between the end of neoadjuvant chemotherapy and surgery, with paired biopsies; co-primary endpoints are the change in Ki67 proliferation index and in proliferation gene expression for the DNA damage response cohorts, the change in CD8-positive stromal TILs and the interferon-gamma signature for the durvalumab cohort, and plasma ctDNA change for the later cohorts. It opened in October 2019 with primary completion expected in December 2026. No results were indexed in September 2026.
