SEVILLA
SEVILLA is a French-British randomised trial comparing ivonescimab, an antibody that blocks both PD-1 and the blood vessel signal VEGF, with FOLFOX chemotherapy as second-line treatment for bile duct and gallbladder cancer. University College London is the UK site.
Overview
SEVILLA (NCT06529718) is a randomised phase 2 run by UNICANCER at 42 sites in France and the United Kingdom, open since October 2025 with 72 patients planned and primary completion listed for August 2027. Eligible patients have histologically proven intrahepatic, perihilar or distal cholangiocarcinoma or gallbladder carcinoma (ampullary carcinoma excluded) that has progressed after first-line treatment, and are randomised to the PD-1 and VEGF bispecific antibody ivonescimab or to FOLFOX, the ABC-06 standard. The primary endpoint is progression-free survival. It is one of the first trials to test a bispecific immunotherapy against chemotherapy in second-line biliary cancer and gives UK patients with gallbladder cancer a randomised second-line option.
