DNAtrix developed viruses, based on a common cold virus, that are engineered to infect and kill cancer cells. Its main candidate, DNX-2401, was tested in brain tumours including glioblastoma and a childhood brainstem tumour. The company has announced nothing since 2021 and its website no longer works.
DNAtrix described itself in 2021 as a privately held company developing virus-driven immunotherapies for cancer, based on an adenovirus engineered to infect and kill cancer cells selectively. DNX-2401 (formerly Delta-24-RGD-4C) was its lead candidate; the company then said it would enter a global pivotal phase 3 study in recurrent glioblastoma, and that it held FDA Fast Track and Rare Pediatric Disease designations for diffuse intrinsic pontine glioma, where it was being tested in a phase 1 study. DNX-2440 is an adenovirus expressing OX40 ligand, first dosed in March 2021 in people with resectable colorectal liver metastases. On ClinicalTrials.gov, DNAtrix-sponsored studies of DNX-2401 in recurrent malignant glioma, with pembrolizumab, and with interferon gamma are completed; its DNX-2440 liver metastasis study (NCT04714983) is suspended, with the reason given as "DNAtrix management decision not based on study design, safety, or site performance". An MD Anderson phase 1 trial of DNX-2401 delivered in mesenchymal stem cells for recurrent high-grade glioma (NCT03896568) is listed as recruiting.
