Duke Street Bio makes DSB2455, a tablet drug that blocks PARP1, a DNA repair enzyme some cancers rely on, while sparing a related enzyme linked to blood-count side effects. It is in an early trial for advanced cancers with DNA repair defects, including cancer that has spread to the brain.
Duke Street Bio Ltd, at 2 Duke Street, London, develops small molecules against DNA damage repair deficiencies in cancer and immuno-oncology targets; it emerged from stealth in May 2022 discussing PARP1 and PARP7 inhibitor programmes. Its development candidate DSB2455 is a brain-penetrant PARP1 inhibitor that the company says is highly selective over PARP2, whose inhibition is linked to blood toxicity with first-generation PARP inhibitors. The European Medicines Agency approved a clinical trial in September 2024, the first patient was dosed in a phase 1 trial in December 2024, and in June 2025 the FDA granted Fast Track designation for patients with homologous recombination deficient brain metastases from triple-negative breast cancer. The registry lists breast, ovarian, metastatic castration-resistant prostate and pancreatic cancers and brain metastases with homologous recombination deficiency. Its first registered cancer trial, NCT06458712, was posted on ClinicalTrials.gov in June 2024.
