Nimbus Therapeutics is a privately held Boston biotechnology company that designs small molecules against difficult targets using computational chemistry. Its lead oncology programme is NDI-219216, an inhibitor of the WRN helicase, which the company lists as being in a phase 1/2 trial in microsatellite instability-high (MSI-H) solid tumours; WRN dependence in MSI-H cancer is a synthetic lethal relationship. The pipeline also includes a preclinical SIK2 immunology programme and metabolic-disease collaborations with Eli Lilly. Nimbus sold its TYK2 inhibitor (now zasocitinib) to Takeda in a deal reported by STAT at 4 billion US dollars upfront, and its ACC inhibitor programme to Gilead. It raised 125 million US dollars in 2022 and 210 million US dollars in 2023 (led by GV with SR One and Atlas Venture), according to trade press coverage of the announcements.
