Relay Therapeutics
Relay Therapeutics uses computer simulations of how proteins move to design precise cancer pills. Its lead drug, zovegalisib, targets a mutated growth protein called PI3K-alpha in breast cancer while sparing the normal version.

Relay Therapeutics is a Cambridge, Massachusetts precision medicine company that combines computational modelling of protein motion with experimental chemistry. Its lead programme, zovegalisib (RLY-2608), is a mutant-selective, allosteric PI3K-alpha inhibitor that the company lists in phase 3 for PIK3CA-mutant breast cancer and in a phase 1/2 trial for vascular anomalies. RLY-8161, an NRAS inhibitor, is in a phase 1/2 trial in NRAS-mutant melanoma and other NRAS-mutant solid tumours. Lirafugratinib (RLY-4008), an FGFR2-selective inhibitor developed in FGFR2-fusion cholangiocarcinoma, was out-licensed to Elevar Therapeutics in December 2024. A non-oncology alpha-galactosidase A programme for Fabry disease is also listed. Relay completed its Nasdaq IPO in July 2020, selling 20 million shares at 20 US dollars per share according to its final prospectus.
