Siren Biotechnology uses a harmless virus to deliver a gene for an immune-signalling protein directly into brain tumours, so the tumour itself makes the immune signal that helps fight it.
Siren Biotechnology, headquartered in San Francisco and founded by Nicole Paulk, develops universal AAV immuno-gene therapy, combining adeno-associated virus gene delivery with cytokine immunotherapy for solid tumours, starting with brain cancer. Its lead SRN-101 is an AAV vector encoding human interferon beta, given as a single intratumoral infusion by MRI-guided convection-enhanced delivery. The FDA cleared its first IND in January 2026, after granting orphan drug and rare paediatric disease designations; the phase 1/2 ALERT-1 trial in adults with recurrent high-grade glioma is not yet recruiting (NCT07902037). The company reports research funding from the California Institute for Regenerative Medicine. Its first registered cancer trial, NCT07902037, was posted on ClinicalTrials.gov on 28 October 2026.
