Tagworks Pharmaceuticals makes antibody-drug conjugates whose cancer-killing payload is released only when a second, separately injected molecule reaches the tumour. Its first product, TGW101, is in an early trial in advanced solid tumours.
Tagworks Pharmaceuticals BV is a Nijmegen, Netherlands, company with operations in the United States that pioneered Click-to-Release, a bioorthogonal chemistry in which a click reaction in the body cleaves a linker to release or deactivate a payload. Co-founded by Marc Robillard and Raffaella Rossin, it builds on click chemistry work recognised by the 2022 Nobel Prize and on the work of Joseph Fox at the University of Delaware. The approach lets antibody-drug conjugates act on non-internalising targets. TGW101 combines a TAG-72-binding diabody conjugated through a TCO linker to the MMAE payload with a separately dosed small-molecule trigger (TRG001) that releases MMAE in the tumour microenvironment. TGW211 is a HER2-directed radioimmunoconjugate whose circulating radioactivity can be cleared after tumour uptake; its CleavHER phase 0/1 imaging study is sponsored by Radboud University Medical Center. Its first registered cancer trial, NCT06959706, a first-in-human phase 1 dose-escalation study of TGW101 in advanced solid tumours at US sites, was posted on ClinicalTrials.gov in May 2025 after FDA clearance of the IND in April 2025.
