EIS-12656

EIS-12656 is a small molecule (oral) from Eisbach Bio, with The University of Texas MD Anderson Cancer Center as trial site. An oral, brain-penetrant small molecule that inhibits the chromatin helicase ALC1 (CHD1L) through an allosteric mechanism. Tumours with homologous recombination deficiency depend on ALC1 to repair DNA damage, so blocking it is designed to be synthetically lethal in them. Its furthest stage is phase 1, for homologous recombination deficient advanced solid tumours, breast cancer and ovarian cancer. ClinicalTrials.gov registers it in one study linked below. Phase 1/2 MATCH trial NCT06525298 at MD Anderson recruiting (registry checked 3 Nov 2026); first patient dosed September 2024. Stage set to phase-1: the only trial is phase 1/2 and no phase 2 portion is reported (same rule used across this batch). Whether it works in people has not been established unless a trial below reports it.
