Glasdegib
Glasdegib is a hedgehog-pathway pill that, with low-dose chemotherapy, extends survival in older AML patients who cannot have intensive treatment; it has largely been displaced by venetoclax combinations.
BRIGHT AML 1003: OS 8.8 vs 4.9 months with low-dose cytarabine. Approved 2018; the BRIGHT AML 1019 trials with intensive or azacitidine backbones were negative, and venetoclax-azacitidine became the preferred unfit-AML regimen.
