Glecirasib
Glecirasib is Jacobio's KRAS G12C inhibitor, approved in China in 2024 for previously treated non-small cell lung cancer with that mutation, and in a phase 3 trial against docetaxel.
Overview
Jacobio Pharma developed glecirasib (JAB-21822) and partnered it with Allist Pharmaceuticals for China. The NMPA approved it in 2024 for KRAS G12C-mutant non-small cell lung cancer after at least one prior systemic therapy, on a single-arm phase 2 trial with a response rate in the range of the approved Western inhibitors and a low rate of gastrointestinal side effects. A randomised phase 3 trial against docetaxel and combination studies with a SHP2 inhibitor are registered.
Binds the mutant cysteine of KRAS G12C and locks the protein in its inactive state, the same mechanism as sotorasib and adagrasib. Connects to KRAS.
