A method that reads whether a breast cancer still carries the oestrogen receptor from the DNA in a blood sample, when there is enough tumour DNA there.
Griffin profiles nucleosome protection and accessibility from cell-free DNA at as low as 0.1x whole-genome coverage, with a GC correction tailored to fragment size. It predicted oestrogen receptor subtype in 139 patients with metastatic breast cancer with at least 5 per cent detectable ctDNA, with an area under the curve of 0.89, and 0.96 in independent cohorts.
The phenotype of advanced breast cancer, not only its mutations, can be read from blood. The method needs percent-level tumour DNA, so it does not yet reach residual disease after surgery.
