CD96
CD96 sits on T and NK cells and binds CD155, the same molecule TIGIT reads on tumour cells. It is the least tested of that family; one blocking antibody is in phase 2 platform trials in lung cancer.
Overview
CD96 (chromosome 3q13.13) may mediate adhesive interactions of activated T and NK cells in the late phase of the immune response, promoting NK-cell adhesion to targets through PVR (CD155) on the target cell; it is expressed on normal T-cell lines and clones and at very low levels on activated B cells (UniProt P40200). CD155 is the ligand for the co-stimulatory receptor CD226 and the co-inhibitory receptors TIGIT and CD96 (Kučan Brlić et al. 2017). GSK6097608, an anti-CD96 antibody, is being tested in phase 2 platform studies with dostarlimab in non-small-cell lung cancer (NCT05565378, NCT03739710).
